摘要 | Hydrogenase plays the crucial role in H2 evolving of microalgae.In this paper, the cDNA of Chlamydomonas reinhardtii CC-124 encoding hydrogenase (hydA1) was obtained from the total RNA by RT-PCR according to the methods described previously (Zhang et al., 2005; Happe and Kaminski, 2002; Forestier et al., 2003). Its ORF profileswere analyzed with biological softwares. Results show that 8 amino acids are replaced in HydA1 of C. reinhardtii CC-124 compared with that of C. reinhardtii CC-503. All the replacements happen at the non-conserved residues except for one at residue 297 which is reported to be necessary for the formation of the Hcluster (Happe and Kaminski, 2002). The replacements between polar amino acids and non-polar amino acids are found at four positions. Codon usage analysis shows that the triplets with C/G at the third position are much more frequently used in algal hydA genes. GC contents of algal hydrogenase ORFs (56.2–68.4%) are much higher than those of Clostridium strains (32.1–34.3%) (Meyer and Gagnon, 1991; Morimoto et al., 2005), which indicates that high GC pressure accidents might happen during the evolution of algal hydrogenases. |
